Virus Inactivation by Formaldehyde and Common Lysis Buffers

Numerous mammalian viruses are routinely analyzed in clinical diagnostic laboratories around the globe or serve as indispensable model systems viral research. Potentially infectious entities handled blood, biopsies, cell and tissue culture samples. Countless protocols describe methods for virus fixation inactivation, yet many, a formal proof of safety completeness inactivation remains to be shown. While modern nucleic acid extraction work quite effectively, data largely lacking on possible residual infectivity, e.g., when assessed after extended times, which maximizes sensitivity low levels infectiousness. Therefore, we examined potency procedures virus-containing specimens applying commonly used fixatives like formaldehyde extraction/lysis buffers. Typical representatives different classes, including RNA DNA viruses, enveloped non-enveloped, such adenovirus, enterovirus, lentivirus, coronavirus, were used, reduction vitro infectiousness was standard protocols. Overall, 30-minute incubation with at room temperature effectively inactivated all tested non-enveloped viruses. Full HIV-1 ECHO-11 also achieved buffers test, whereas SARS-CoV-2 AdV-5, only five seven lysis fully effective under conditions.
